Yes, you leave the glow plugs wired up. That’s why you can use the harness at the relay instead of having to fish around at the engine block with your meter. Also ensures you know you don’t have a bad connection.

IIRC good plugs are around 0.6-0.8 Ohm. You’re really looking at differences between them to flag where there may be an issue and investigate further.
